A whole blood filtering method and a filter membrane structure for whole blood filtering. The method specifically comprises the following steps: (1) selecting a filter membrane structure formed by stacking at least two layers of filter membranes from top to bottom in sequence, and performing red blood cell agglutinin treatment on the filter membrane structure for future use; (2) adding a whole blood sample to the filter membrane structure for filtration; and (3) collecting the filtered serum or plasma. The filter membrane structure is formed by stacking at least two layers of filter membranes from top to bottom, and pore diameters of the stacked filter membranes gradually decrease from top to bottom and the areas gradually increase or are equal. The filter membrane structure is formed by stacking at least two layers of filter membranes, and red blood cell agglutinin treatment is performed on the filter membrane structure, so that red blood cells in the whole blood sample are combined with the red blood cell agglutinin in the filter membranes and are trapped in the filter membrane structure, thereby ensuring that the red blood cells are completely filtered and adsorbed in the filter membrane structure, and therefore, plasma/serum can be effectively and stably separated from a very small amount of blood.
