The present invention relates to a novel mutant 5′-inosinic acid dehydrogenase capable of producing 5′-inosinic acid of high yield, and to a method for preparing 5′-inosinic acid using the novel mutant 5′-inosinic acid dehydrogenase. The novel mutant 5′-inosinic acid dehydrogenase is prepared by i) substituting threonine for a 377^th amino acid, ii) substituting isoleucine for a 499^th amino acid, or iii) substituting threonine for a 377^th amino acid and substituting isoleucine for a 499^th amino acid, in the amino acid sequence of SEQ ID NO: 2.
