The present invention includes a first pair of acousto-optic deflectors (x-axis deflectors) that deflect the laser beam in the xz plane to focus the laser beam in 3D and the laser beam in the yz plane. Using a 3D laser scanning microscope with a second pair of acousto-optic deflectors (y-axis deflectors) deflected at a substantially straight line (in a given direction and in any direction in 3D space) 3D line). The invention further provides for focusing the laser beam within a 3D space defined by the optical axis (Z) of the microscope used and the X and Y axes perpendicular to the optical axis and perpendicular to each other. It also relates to a method of scanning a region of interest with a 3D laser scanning microscope equipped with an acousto-optic deflector.
