The present invention relates to a PCL and PEG patch tissue regeneration support, and a manufacturing method thereof. More specifically, the manufacturing method comprises: a step of preparing a polycaprolactone-polyethylene glycol copolymer solution by adding polycaprolactone (PCL) and polyethylene glycol (PEG) and acid to an organic solvent and by stirring the same; a step of preparing an electrospinning solution by adding a growth factor to the solution and by stirring the same; and a step of collecting nanofibers arranged in a spindle form on a collector by operating an electrospinning device after connecting the prepared electrospinning solution to a pump. The PCL and PEG patch tissue regeneration support according to the present invention can be usefully used for treatment of acute and chronic tympanic membrane perforation, and in particular, can reduce the proportion of surgeries and show high tympanic recovery rates in a therapy depending on surgeries for chronic eardrum perforation treatment. In addition, the PCL and PEG patch tissue regeneration support can be applied to regeneration and treatment of damaged other tissues, and can develop and apply various tissue regeneration supports by imitating tissue structures of other parts of a human body.
