The present invention relates to a species-specific primer set for discriminating species of two skin webfoot octopus, one skin webfoot octopus, long arm octopus, and Japanese flying squid, and a high-speed analysis method of gene amplification. Mixing of two skin webfoot octopus into a raw material for cooking which includes one skin webfoot octopus can be prevented by the primer set or high-speed analysis method of gene amplification using the same. Additionally, species of one skin webfoot octopus can be easily guaranteed, and proper pricing of one skin webfoot octopus which is domestic webfoot octopus, long arm octopus, and Japanese flying squid is possible. Moreover, the present invention is helpful for revitalization of distribution and a consumer market by exposing an example that two skin webfoot octopus which is imported webfoot octopus is disguised into domestic webfoot octopus, long arm octopus, squid, or the like, and monitoring imported webfoot octopus.
