A method for the detection of human antibodies directed against alloantigens human platelets (HPA) in a sample, comprising the steps: (a) providing at least one surface having at least human platelets or membrane fractions of platelets attached to the thereof, wherein the HPA of interest are present in said platelets or platelet membrane fractions are known; (B) contacting said surface with said sample, wherein human antibodies directed against HPA which are present in said platelets or membrane fractions of platelets contained in said sample are immobilized on said surface specifically binding to platelets or platelet membrane fractions from step (a); (C) contacting said surface with a detection agent fluorescently labeled, wherein said detection agent specifically binds to human antibodies directed against HPA which are present in said platelets or membrane fractions of platelets, and the said detection agent respective human antibodies contained in said sample, and human platelets or platelet membrane fractions from step (a) form a complex bound to the surface; (D) exciting said complex attached to the surface with an evanescence field of a light source; (E) measuring the fluorescence emitted from said complex attached to the surface; and (f) detecting human antibodies against HPA which are present in said platelets or membrane fractions of platelets based on the emitted fluorescence measured in step (e), where platelets or membrane fractions of platelets are platelets or plaque fragments comprising native glycoproteins.
