A method for detecting at least one alloantigen human platelets (HPA) in human platelets contained in a sample, which is anti-coagulated whole blood in a concentration of more than 25%, comprising the steps of: (a) providing at least one surface having at least one antibody bound thereto, wherein he said antibody is directed against the glycoprotein carrying at least one HPA is detected; (B) lysing the platelets contained in the sample and solubilize membrane proteins of said platelets; (C) contacting said surface with the lysed sample solubilized step (b), wherein the respective glycoproteins contained in said sample are immobilized on said surface by specifically binding to the antibody of step (a); (D) contacting said surface with a fluorescently labeled antibody, wherein said antibody is directed against at least one HPA is detected, and wherein said antibody, the respective glycoproteins contained in said sample, and antibody step (a) form a complex bound to the surface; (E) exciting said complex attached to the surface with an evanescence field of a light source; (F) measuring the fluorescence emitted from said complex attached to the surface; and (g) detecting at least one HPA based on the emitted fluorescence measured in step (f).
