A method for preparing variants (Y') of a protein (Y) that have improved binding properties for a target molecule (X), is new. A method for preparing variants (Y') of a protein (Y), comprises: (a) preparing an in vitro expression system consisting of a nucleic acid sequence (S), encoding Y; a target molecule (X) able to bind to Y and Y'; RNA polymerase (Pol), able to transcribe S; and a reverse transcriptase (RT) able to transcribe the transcript from S, where either X is coupled to Pol and Y to RT; or X is coupled to RT and Y to Pol; (b) incubating the system under conditions that: (a) allow transcription, reverse transcription and translation, to generate Y' (and corresponding coding sequences S'); and (b) favor formation of Y' that have improved binding for X; and (c) isolating Y' with improved binding for X and/or isolation of the corresponding coding sequence S'. An independent claim is also included for a kit for the process.
