Described herein are tools and a method for identifying oioid ligands (agonists or antagonists) that bind and (in)activate mammalian Opiod reseptors. The method is based on the use of stably transformed lepidopteeran cell lines that over-express mammalian opioid receptors. In some cell lines, the promiscuous Caq protein, Ga16, is also over-expressed to enhance the coupling of the over-expressed opioid receptors to the Ca2= signaling pathway. Opioid activity is monitored through the measurementsof intrecellular Ca2= release by fluorescent or luminescent methods.
