Method for producing full-length cDNA (I). Method for producing full-length cDNA (I) comprises: (a) dephosphorylation of nucleic acid fragments that do not have a 5'-cap; (b) removing the 5'-cap from capped RNA; (c) ligating an oligonucleotide (ON) to the 5'-end of uncapped RNA; and (d) synthesis of (I) by reverse transcription from the RNA of (c). The new feature is that step (c) uses a ligase and a double-stranded adapter (X) with cohesive ends. Independent claims are also included for the following: (1) method for ligating (X) to the 5'-end of an RNA; and (2) kit for preparing (I).
