In the present invention, there is disclosed a process for purifying double-stranded DNA wherein the process is characterized in that it comprises at least one purification stage in which a solution containing said double-stranded DNA in a mixture with other components is passed over a support on which is covalently coupled an oligonucleotide capable of hybridizing with a specific sequence present on said DNA to form a triple helix wherein said specific sequence is represented by a sequence being naturally present in the double-stranded DNA or a sequence that has been introduced artificially into the double-stranded DNA. The invention also relates to a purification process of a double-stranded RNA as well as a purification process of a plasmid DNA, wherein both the purification processes are carried out in the same manner.
