A procedure for detecting a thin layer (2) of a biological or chemical material not more than 50 nm thick located on a support (4) which absorbs light in a predetermined area of the visible spectrum close to the infrared end consists of taking one measurement in the absence of the layer, a second when it is present, and comparing the two measurements. The thin layer can be e.g. of metal and a dielectric material, and the support can be a semiconductor, glass or silicon substrate (30). The measurements are made with the aid of a pump laser (6), a photodiode detector (10) and a processor (12).
