PURPOSE: Provided is a GIGANTEA gene which regulates biological clock and blossom of Arabidopsis thaliana. And its separation method is also provided. The GIGANTEA gene is very useful in genetic engineering, especially when the gene is used in cultivating crops, it brings economic benefits. CONSTITUTION: GIGANTEA gene of Arabidopsis thaliana is separated by Map-Based Cloning method using Arabidopsis thaliana requiring 1 gene(TH1), to use itself for probing a regulating gene of biological clock or blossom of Arabidopsis thaliana, and for improving biological clock related traits. The GIGANTEA gene separation is characterized by the steps of: i) obtaining a filter made of BAC(bacterial artificial chromosome) libraries and two clones, T14K14 and T22J18, from the filter through a general library screening method; ii) implementing RFLP(restriction fragment length polymorphism) analysis to figure out the difference of cleaved pattern of wild type and mutant DNA by cutting DNA separated from gi-1 and gi-2 with restriction enzymes, electrophoresing DNA segments, and conducting southern blotting; and iii) performing PCR by using T22J28 as a template with LA PCR DNA polymerase to obtain 9kb segment and in the same way obtain 5kb segment.
