A procedure for producing recombinant -galactosidase from Thermus sp. (strain T2) in host cells, and for purifying it in a single chromatographic step. Completely new nucleotide sequence which corresponds to a gene of Thermus sp. (strain T2 ATCC 27737) and which codes for a heat-stable -galactosidase. Production of gene constructs for the high-level expression of said nucleotide sequence in Escherichia coli cells by cloning them in expression vectors. Purification of a hybrid protein with heat-stable -galactosidase activity produced in E. coli in a single chromatographic step. This heat-stable chimeric protein has application in hydrolysis of milk and milk products, reduction of the lactose content of whey effluent, cold labelling of nucleic acid probes by conjugation therewith, synthesis of oligosaccharides by transglycosylation and rapid evaluation of moderate heat treatments.
