The revelation plasmid of the human thyroid per oxydase, the transformation CHO cell, the anti- microsome antibody measurement kit null which includes the human thyroid per oxydase and the said human thyroid per oxydase which are produced genetic engineering
PURPOSE:To determine an anti-microsome antibody in high sensitivity by adopting human thyroid gland peroxidase derived from cultivation of transformant with a specific manifestation vector as using an antigen and determining reactivity of said antigen with a blood serum. CONSTITUTION:A vector inserted of clonized human thyroid gland peroxidase gene is cut with a restriction enzyme to obtain a DNA fragment containing human thyroid gland peroxidase (hTPO) gene. On the other hand, a manifestation vector in another animal cell or Escherichia coli is cut with a restriction enzyme to obtain a DNA fragment containing a promoter region, a terminator region and a replication-initiating point, then said DNA fragments are jointed to a re-construct vector. Thus, Escherichia coli, etc., is transformed with the resultant vector and a transformant is cultured to produce hTPO, then said hTPO is adopted as using an antigen, thus reactivity with a blood serum is determined to determine an anti-microsome antibody.
