1320056 Opium alkaloid antigens and antibodies F HOFFMANN-LA ROCHE & CO AG 13 May 1971 [13 May 1970] 14629/71 Heading A5B [Also in Divisions B1 and C2] An opium alkaloid antigen consisting of a carboxy-C 1-6 alkyl derivative of the phenolic hydroxy group of an opium alkaloid coupled to an immunogenic carrier material is prepared by reacting the opium alkaloid derivative with an immunogenic carrier material. The immunogenic carrier material may be a protein such as human gamma globulin, human serum albumin, rabbit serum albumin, bovine gamma globulin and bovine serum albumin; a natural or synthetic polymeric compound such as a polypeptide e.g. polylysine or a polysaccharide. The opium alkaloid may be morphine, heroin, codeine or nalorphine or a derivative thereof and the preferred carboxyalkyl derivative of an opium alkaloid is 3-O-carboxymethylmorphine. The reaction may be carried out in an aqueous reaction medium having a pH of 3 to 6.5, at 0‹C to 50‹C, using a molar excess of 30 to 10,000 of the opium alkaloid derivative, in the presence of a water-soluble coupling agent such as a carbodiimide e.g. 1-ethyl- 3-(3-dimethylaminopropyl) carbodiimide. An opium alkaloid specific antibody consisting of a gamma globulin fraction protein having a multiplicity of sites which will selectively complex with an opium alkaloid, a carboxyalkyl derivative of the phenolic hydroxy group of an opium alkaloid or an opium alkaloid antigen consisting of a carboxyalkyl derivative of the phenolic hydroxy group of the opium alkaloid coupled to an immunogenic carrier material is prepared by injecting the opium alkaloid antigen into a host animal repeatedly over a period of time until opium alkaloid specific antibody is present in the blood stream, collecting a blood sample from the host animal after the injection period is completed, separating the serium from the blood sample, precipitating the antibodies from the serum by treating the serum with a neutral salt solution e.g. ammonium sulphate and purifying the antibodies.
