Raman systems and methods for detecting tumor marginal cancer tissue in real time during a surgical procedure are provided. The system includes a laser excitation source, a probe, a spectrometer, and a camera specially designed to use Raman spectroscopy in real time during surgery. The methods of the invention relate to quality assessment of Raman data, comparison of the obtained Raman data with known biomarkers, and tissue classification (cancerous or non-cancerous) based on the corresponding Raman data. Systems and methods enable the detection and classification of tissues using Raman spectroscopy in previously unavailable, real-time surgical applications, including the identification of cancer subtypes.
