    • 摘要: Impaired axonal transport has been observed in patients with amyotrophic lateral sclerosis(ALS)and in animal models,suggesting that transport proteins likely play a critical role in the pathological mechanism of ALS.Dysregulation of Kinesin-family-member 5α(Kif5α),a neuron-specific isoform of heavy chain kinesin family,has been described in several neurological disorders,in humans and animal models,including ALS.In this study,we determined Kif5αexpression by gene sequencing,quantitative reverse transcription-polymerase chain reaction,and western blot assay in the cervical spinal cord of wobbler mice and immunofluorescence staining in dissociated cultures of the ventral horn.Further,we observed the distribution of Kif5αand mitochondria along motor neuronal branches by confocal imaging.Our results showed that Kif5αexpression was greatly dysregulated in wobbler mice,which resulted in altered distribution of Kif5αalong motor neuronal branches with an abnormal mitochondrial distribution.Thus,our results indicate that dysregulation of Kif5 and therefore abnormal transport in motor neuronal branches in this ALS model could be causative for several pathological findings at the cellular level,like misallocation of cytoskeletal proteins or organelles like mitochondria.

    • 摘要: An experimental unit was built which was located in the vicinity of the domestic wastewater treatment plant of the Municipality of San Marcos in the Department of Carazo. Said unit consisted of two welded metal barrels with a total height of 1.8 m and a diameter of 0.90 m, likewise, this unit comprised three functional zones;the first or input area composed of a collection unit, a driving unit, a flow control unit, and two distribution units;the second or packed zone, which is the axis of the system because it is there where bacterial growth takes place and where the removal of organic matter takes place;and an outlet composed of PVC pipe [1]. The main objective of this work was to determine how efficient a rocky filter turns out to be, using stabilization pond effluent as a post-treatment unit using red concrete as a filter medium. The study consisted in carrying out laboratory tests on water samples taken from the influent and effluent of a filter made up of red rock, as a filter medium, in order to determine the percentage of organic matter removal efficiency that it has used a hydraulic retention time of 19 hours, 15 hours, and 12 hours [2]. Among the aspects to be considered in the experimental study was the characterization of the effluent from the treatment system of San Marco, Carazo, to determine the quality of water that will be of interest to the study [3]. In this stage, four composite samplings were carried out, two of 12 hours and two of 24 hours, in which physical, chemical, and microbiological parameters were analyzed. Likewise, to determine the efficiency of the filter, the same parameters considered in the characterization of the lagoon effluent were analyzed, obtaining removal efficiencies of up to 55.53% in organic matter, and 64.60% in suspended solids for a hydraulic retention time of 19 hours;In the same way, the efficiency of the filter was carried out for retention times of 15 hours and 12 hours, in which organic matter removals of 67.84% and 56.59% and 82.77% and 77.51% in total suspended solids were found, respectively.

    • 摘要: A practical experimental method is proposed to investigate thermal transport by characterizing the motion of plasma flows through a x-ray spectroscopic technique using tracers.By simultaneously measuring multiple parameters,namely,the mass-ablation rate,the temporal evolution of plasma flow velocities and trajectories and the temperature,it is possible to observe a variety of physical processes,such as shock wave compression,heating by thermal waves,and plasma thermal expansion,and to determine their relative importance in different phases during the irradiation of CH and Au targets.From a comparison with hydrodynamic simulations,we find significant differences in the motion of the plasma flows between CH and Au,which can be attributed to different sensitivities to the thermal transport process.There are also differences in the ablation and electron temperature histories of the two materials.These results confirm that velocities and trajectories of plasma motion can provide useful evidence in the investigation of thermal conduction,and the approach presented here deserves more attention in the context of inertial confinement fusion and high-energy-density physics.

    • 摘要: Chito-oligosaccharide(COS)had shown a variety of biological activities and potential biomedical implications.The present study investigated the pharmacokinetics,bioavailability,and in vitro absorption of COS with degrees of polymerization(DPs)2-7 and explored the influence of DPs on them.From Caco-2 cell permeation studies,COS were low permeability compounds with no directional effects,suggesting a low in vivo absorption mediated by facilitation diffusion and paracellular absorption.After an intragastrical administration to rats,COS2 showed the highest systemic exposure in six oligosaccharides.The bioavailability of COS2-7 was 7.33%,6.11%,4.67%,4.13%,4.02%,0.99%,respectively.Differences in bioavailability for each COS correlated to structural variations,with high DPs contributing to a decrease in bioavailability.In conclusion,COS could be absorbed by the intestinal tract both in vitro and in vivo.The very low oral bioavailability of COS could be due to low permeability.DPs can affect absorption and bioavailability of COS2-7.This study provided evidence for the absorption characteristics of COS2-7 to help us better understanding the pharmacological actions.

    • 摘要: [Objectives]To explore the differences in nitrogen accumulation and transport characteristics and grain protein content of wheat varieties with different nitrogen efficiencies and their responses to irrigation.[Methods]Under field conditions,using nitrogen-inefficient varieties Luohan 17 and Xinhua 818 and nitrogen-efficient varieties Bainong 418 and Bainong 419 as materials,this paper studied the nitrogen accumulation and transport characteristics,grain protein content and protein yield of wheat with different nitrogen efficiencies under rainfed and irrigated conditions.[Results]Compared with the nitrogen-inefficient wheat varieties,the pre-flowering nitrogen transport and the shoot nitrogen accumulation at the mature stage of nitrogen-efficient wheat varieties decreased by 15.08%and 28.25%,respectively,and the grain protein content decreased by 11.66%,under rainfed conditions.Compared with rainfed conditions,nitrogen accumulation in shoots of nitrogen-inefficient wheat varieties and nitrogen-efficient wheat varieties at the mature stage increased by 6.59%and 67.05%,respectively,and grain protein content decreased by 13.50%and 3.47%,respectively,under irrigated conditions.The two nitrogen efficiency types of wheat had different responses to irrigation after flowering.After irrigation,the nitrogen accumulation of nitrogen-efficient varieties increased by 274.80%,while that of nitrogen-inefficient varieties decreased by 51.15%.Finally,the grain protein yield of nitrogen-inefficient wheat varieties remained stable,while the grain protein yield of nitrogen-efficient wheat varieties increased by 40.37%.[Conclusions]The nitrogen accumulation and transport characteristics and grain protein content of wheat varieties with different nitrogen efficiencies are different under different irrigation conditions.In production,it is necessary to take different irrigation measures in accordance with the difference in nitrogen efficiency of wheat varieties,so as to increase the protein content of wheat grains.

    • 摘要: Objective:To reform the transfer process of surgical patients,standardize the transfer process,and improve the quality of safe transfer;unify the intrahospital transfer process to achieve standardized management;ensure patient safety,improve work efficiency,and reduce the occurrence of adverse events.Methods:A transfer team was formed,a handover record sheet for surgical patients was designed,and the workflow and specifications for handover and transfer of surgical patients were formulated.Results:After using the re-designed handover record sheet for surgical patients and improving the procedure for transferring surgical patients,the accuracy of surgical patient handover was ensured,the handover and transfer time was shortened,and the surgical turnover rate improved.Conclusion:By designing and reforming the service model,it is possible to change and standardize the transfer process of surgical patients and improve the quality of safe transfer,the operating room management system so that it is more rational and humanized,the management model,the working environment,as well as the overall quality and efficiency of operating room nursing.

    • 摘要: This paper outlines research that explores a range of digitally-driven opportunities and challenges in seeking to get cities moving and functioning better;with a focus on the application of Artificial Intelligence and Blockchain technologies.The paper identifies specific potential applications and outlines where such technology can provide tangible benefits for the transport sector and the associated policy and management structures.Specific benefits explored include cost savings(such as digital payment mechanisms,virtual toll collection and congestion charging,capital deferment through greater network utilisation);time savings(from improved transport network operation and management processes);improved operations(such as real-time pay-as-you-drive systems with network differential pricing ability);improved safety(from less collisions and reduced impacts);improved user experience(such as improved platforms and automated payments for parking and bookings);and improved freight and logistics(such as secure storage of time-stamped travel-related data).The paper presents a set of industry partner-preferred use cases such as congestion management,road user pricing,asset management,mobility-as-a-service,and freight logistics.The paper outlines specific opportunities for Artificial Intelligence and Blockchain technology to provide value to transport agencies and service providers either individually or in combination.The paper highlights risks and rewards for the transport sector,including a further review of early findings from the application of such technologies in transport sectors around the world.This research has been developed with funding and support provided by Australia’s Sustainable Built Environment National Research Centre(SBEnrc)and its partners,and has been presented at the CIB World Building Congress 2019.
