In the rat heart, we have shown that chronic deficiency of nigrostriatal dopamine resulted in impaired systolic and diastolic function and increased left ventricular myocardial stiffness. A decrease in cardiac contractile function resulted from an increase in an amplitude of swelling of the isolated cardiac mitochondria to calcium in physiological concentrations and calcium overload, which gives evidence of mitochondrial permeability transition pore (mPTP) opening. Increased permeability of the mitochondrial membrane was associated with elevated mitochondrial reactive oxide species (ROS). Administration of ROS scavenger and mPTP inhibitor melatonin one hour prior an experiment decreased significantly mitochondrial permeability and restored cardiac contractile function in the rat heart with chronic deficiency of nigrostriatal dopamine
