Phenlyalanine ammonia-lyase (PAL) catalyzes the first step in the phenylpropanoid pathway and plays an important role in the biosynthesis of phenylpropanoid and flavonoid compounds. Cis-elements of the promoter region of g2bPAL gene, a PAL gene obtained from popular, Populus kitakamiensis, were analyzed by transient expression in carrot protoplasts and transgenic tobacco. Four conserved cis-elements, M-, G-, L- and P'-boxes in the PAL promoters of several plant species, were found in the proximal region of the g2bPAL promoter. Deletion of G-box in the promoter resulted in the loss of g2bPAL promoter activity in carrot protoplasts. In transgenic tobacco plants harboring g2bPAL-GUS, deletion of P'-box resulted in strong GUS expression in the endodermis of the stems and in the root. The findings indicate that the P'-box may play an important role in the regulation of g2bPAL expression to synthesize lignin during xylem development.
