The rapid detection, identification and separation of spoilage and brewing yeasts are of serious interest for the quality control of the beer production process. The potential of the PCR-DHPLC (Polymerase Chain Reaction -^sDenaturing High Performance Liquid Chromatography) method was studied to detect the presence of spoilage yeast populations in single and mixed cultures. PCR was performed with universal primers amplifying the ITS 1 and ITS2 rRNA genes of yeast species within a mixed population, followed by DHPLC to separate the different PCR-products. Applying this method, 21 of the 25 yeast species tested were clearly separated from each other, showing the potential of PCR-DHPLC in the quality control process. The other four yeast species formed two clusters. Additionally some Saccharomyces cerevisiae non brewing strains and strains very closely related to brewing yeast strains could be differentiated from brewing yeast strains. Using a Primer pair specific to S. cerevisiae yeasts the PCR-^sDHPLC method showed the potential to differentiate pure brewing culture yeast strains.
