A computational model for the prediction of buccal permeability of a drug using molecular descriptors was developed. A porcine buccal model was used for the in vitro permeation studies of 13 drugs. Multiple linear regression showed that molecular volume, logD_(6.8), number of hydrogen bond donors, and number of rotatable bonds were the most important parameters describing logK_p.
