Randomly amplified Polymorphic DNA (RAPD) fingerprint and spore coat protein in Bacillus thuringiensis and related Bacillus species were investigated. The homogeneity of H-antigen was investigated simultaneously. It was showed that there were no homogeneity of H-antigen in 5 Bacillus species. From 6 arbitrary primers the primer NP2 was the only one which was able to produce PCR products in all tested species. The RAPD fingerprints of various species were different, and those of various subspecies and strains were different also. The SDS-PAGE patterns of spore coat proteins in 5 species showed a distinguished difference, but there were a partial homogeneity between B. thuringiensis and B. cereus, and high similarity in different subspecies of B. thuringiensis. It was proposed that the SDS-PAGE patterns of spore coat protein could be used as a important item for the taxonomy of genus Bacillus and B. thuringiensis.
