Cucumber mosaic virus (CMV), the type member of the Cucumovirus genus, is one of the best-characterized tripartite positive-single stranded viruses. Systemic symptom determinants in CMV genomes have been extensively studied. Among systemic symptoms in virus-infected plants, the yellow/green mosaic symptom in CMV-infected tobacco was shown to be controlled by one amino acid substitution at position 129 of the CMV coat protein However, little is known about the molecular mechanism of coat protein-associated mosaic symptom expression in CMV-infected tobacco. For a better understanding of the molecular basis of mosaic symptom expression in CMV-infected tobacco, further information about the virus determinant for this symptom is needed. In this report, we isolated three CMV mutants from regenerated tobacco plants showing different symptoms from the original yellow mosaic symptoms in the yellow strain of CMV [CMV (Y)] -infected tobaccos, and analyzed their molecular determinants for symptom expression.
