The genus Serpulina includes certain pathogenic intestinal spirochetes and some species of the genus have recently been unified under the genus designation of Brachyspira (B.). B. hyodysenteriae is the agent of swine dysentery (3) and Serpulina (S.) intermedia has been implicated as a cause of typhilitis and colitis in poultry. B. alyinipulli is a cause of diarrhea in chickens and B. pilosicoli is the agent of intestinal spirochaetosis in pigs and also infects a number of other species, including humans, poultry and dogs. B. innocens and B. murdochii are considered nonpathogenic commensal organisms (3). Identification of spirochetes was originally performed on the basis of phenotypic methods (2,3). More recently, a variety of genetic-based methods have been developed, including amplification of DNA sequences by polymerase chain reaction (PCR) assays of sequences of 16S rRNA, 23S rRNA and the nox gene (1,2). These techniques are very useful for typing purposes, but individual amplifications must becarried out to diagnose each spirochetal species. The present study examined reference isolates of swine intestinal spirochetes representing previously established genetic groups using RFLP of a 307-bp sequence of the nox gene obtained by PCR amplification with conserved primers.
