It was found in our previous paper (1,2) that the aqueous dispersions of DL-lactide/glycolide copolymer (PLGA) nanospheres with peptide administered pulmonaryly to guinea pig with a nebulizer improved significantly and prolonged extensively the biological action. In the present study, powdered PLGA nanocapsules and powdered matrices of hydroxypropylmethylcellulose phthalate (HPMCP) nanospheres with calcitonin were developed to improve systemic delivery of the peptide with dry powder inhalation as well as to improve compliance by reducing inhalation time with the nebulizer (e.g. 20min). To improve inhalation efficiency, the micromeritic properties of powdered nanoparticulate system were specifically designed so as to decrease their aerodynamic diameters by increasing the dynamic shape factor and/or porosity.
