We report the inhibition of tumor growth in the syngeneic K1735-M2 murine melanoma model with liposomal doxorubicin (Ldox) targeted to the α_vβ_3 integrin by an RGD mimetic. Single bolus injection of doxorubicin (dox) encapsulated in an α_vβ_3integrin-targeted liposome (ITLdox) resulted in statistically significant inhibition of tumor growth rates in vivo relative to control injections including sucrose, dox, Ldox, and α_vβ_3 integrin-targeted liposomes (ITLs). In vitro, ITLs bind to purified integrin, and ITLdox inhibits the cell proliferation of murine endothelial cells relative to controls, but tumor cells are not affected under identical conditions. These results suggest that ITLdox inhibits tumor angiogenesis by selectively killing tumor endothelial cells.
