Most research on primary sensory neurons pertaining to nociception and pain perception has focused on smaller neurons with thin-caliber axons that are either unmyelinated C fibers or lightly myelinated A8 fibers. A wide variety of C fibers and A8 fibers collectively have preferred stimuli that cover a range of temperatures (thermoreceptors), a variety of exogenous and endogenous molecules (chemoreceptors), intense mechanical impact (high-threshold mechanoreceptors), or some polymodal combination of such stimuli (Burgess and Perl 1973; Fields 1987; Birder and Perl 1994). Extremes in such stimuli are perceived as pain. In general, C-fiber-mediated nociception has been implicated in slow, burning pain, whereas A8-fiber nociception has been implicated in sharp, pricking pain. Under a variety of pathological conditions, pain perception increases acutely or chronically due to reduced thresholds to normally noxious stimuli (hyperalgesia) or to changes that result in normally non-noxious stimuli becoming noxious (allodynia).
