A rapid protocol for preparing genomic DNA from leaf; root and seed was developed for quick screening of breeding populations and single seed genotyping of wheat. A small piece of leaf; root; or seed endosperm was ground and treated with sodium hydroxide and the resultant solution was used as DNA template for PCR amplification of SSR and STS markers. This method was particularly useful for preparing template DNA from leaves of backcrossed plants for early marker assisted selection of these plants.This method was vaHdated in a breeding program for screening a large number of backcrosses with SSR and STS markers. Single seed genotyping was used for validation of an SSR marker on an F2 population. This method was also used to prepare template DNA from a barley population where a large number of SSR markers were screened.
