High protein content and gluten strength in durum (Triticum turgidum L. var durum) are important to produce pasta; couscous; and burghul. The aim of this study was to construct a duram genetic linkage map and to determine QTLs linked to grain-quality. The studied OmrabiS/ Triticum dicoccoides 600545// Omrabi5 population comprises 114 RILs and the markers used are Single Sequence Repeats markers (SSRs); Amplified Fragment Length Polymorphism markers (AFLPs); and seed storage proteins (SSP). Yellowpigment (YP) and protein content (PC) were analyzed over 18 environments using Near-Infra-Red and chemical protocols. The map consisted of 14-dumm chromosomes plus an unknown group. Analysis of QTLs was based on simple and simplified composite intervalmapping (SIM and sCIM). PC was highly affected by environments. Six PC QTLs were detected on 4BL; 3BS; and 6AS explaining 27% of the total variation and showing QTLxE interaction effects; two of them were linked to vitreousness. The QTLs explanation rate was higher in stressed than in non-stressed environments. T. dicoccoides 600545 alleles improved significantly the PC content. For YP; 3 QTLs were detected on the telomeric regions of 7AL and 7BL; explaining 62% of the total variation. The detected QTLs will be used in the marker assisted breeding program.
