A protocol for preparation of suspension of intact chromosomes of duram wheat from synchronized root tips is described. Histograms of fluorescence intensity (flow karyotypes) obtained after the analysis of DAPI-stained chromosomes contain two composite peaks and no single chromosome may be discriminated. However, the use of double ditelosomic lines facilitates sorting of all chromosome arms. Using FISH on flow-sorted chromosomes, genomic distribution of four repetitive DNA sequences have been determined. An unexpected finding of this study is the discovery that 'Langdon' + 'Chinese Spring' telosomic hues may be used to sort any chromosome arm of hexaploid wheat. Thus, the tools are available to fractionate the genomes of wheat to simplify their analysis.
