We have established 16 different colored cell lines of Portulaca sp. cv. 'Jewel' (JR inbred) including 7 lines on modified Murashige and Skoog (mMS) and 9 lines on modified Girod and Zryd (mJl) culture medium. Betalain composition of those cell lineswas determined by using the high performance liquid chromatography -UV-VIS-photodiode array detector (HPLC-UV-VIS-PDA) method. The cell lines were distinctly different on their betalain contents. Betacyanin and betaxanthin, greatly varied in quality andquantity from line to line. N12 mMS-cell line, a magenta color phenotype, contains highest amount (4 87 mg/g FW) of total betacyanin and N12Or mJ1-cell line, an orange color phenotype, contains highest amount (3819 89 mu g/g FW) of total betaxanthin among all the lines. The cells cultured on mMS medium synthesize more betacyanin in comparison with mJ1 medium. In contrast, mJ1 medium cultured cells synthesize more betaxanthin than that of mMS medium. Addition of natural auxin, indole-3-acetic acid (IAA), to both types of culture media instead of synthetic auxin, 2,4-dichloro-phenoxyacetic acid (2,4-D) affected the growth and betalain biosynthesis 6 mu M and 9 mu M IAA significantly increased the betalain content during the first subculture than that ofcontrol in 12 mMS-cell line and 4YmJ1-cell line (yellow color phenotype), respectively. Effects of different light conditions on betalain biosynthesis were also studied with a magenta color and a yellow color cell lines. Continuous blue light treated cells optimistically influenced the betacyanin and betaxanthin contents in both colored cell lines than the cells cultured on continuous white or 16 h/day white light conditions and the rate of cellular betalain contents increased drastically under continuous white and blue light conditions along with the increasing number of growth cycle.
