Increased levels of TGF-beta1 have been identified in prematurely closing cranial sutures in both a murine model and in humans. Therefore, regulating the expression levels of key growth factors or their receptors may provide a non-surgical means of intervention to delay suture fusion. In this study we examine the ability of TGF-beta siRNA to regulate the gene expression of the factors TGF-betal and FGF-2 as well as their receptors TGF-R1, TGF-R2, and FGF-R1 in a mouse dura cell culture. 16nM TGF-beta1 siRNA is capable of reducing TGF-beta1 mRNA expression significantly when harvested 10 days post-transfection, but expression returned to levels found in mock transfected cells by day 14. Similarly, TGF-R2, FGF-2 and FGF-R1 levels showed an initial reduction in expression on day 4. However, all were near control level by day 7.
