Matrix-assisted laser desorption/ionization (MALDI) mass spectrometry is a widely utilized tool for the analysis of biological samples. However, this technique suffers from some drawbacks, such as the background generated by the MALDI matrix ions during the ionization process. Although the background tends not to be a problem for the analysis of large biomolecules (peptides, proteins, and sugars), analysis of smaller biomolecules is more problematic. The background from the matrix ions tends to obscure ions less than m/z 500. An additional drawback when utilizing MALDI is that most matrix recrystallization techniques do not foster uniform crystal formation. Without a uniform matrix layer, sample signal across the layer will correspondingly be irregular. Uneven crystal formation forces scientists to search for a "sweet spot" in the matrix in order to get optimal signal from the sample.
