An Isotope-Free Method for Quantitative Proteomics

Quantitative proteomics is becoming a prevailing technology in biomedical research for discovering biomarkers, uncovering disease pathways, and revealing novel targets for therapeutic development. Current methodologies for protein quantitation include 2DE, stable isotope labeling (e.g. metabolic labeling and ICAT) and chemical derivatization (e.g iTRAQ) strategies. The labeling-based technologies have certain disadvantages, for example, nonspecifically captured peptide background, frequent difficulty in obtaining useful tandem mass spectrometry fragmentation patterns, inability to quantify metabolites , and high cost. New methods without isotope labeling are emerging for quantitative proteomics. We hereby report a simple, sensitive and reliable labeling-free method for quantitative analysis.
