Intact protein mass spectrometry analysis is an attractive alternative to peptide map due to the ease and the speed of the analysis as well as the information this method can reveal without introducing putative modifications in the course of sample preparation. Intact protein LC/MS analysis can be used to obtain not only molecular weight but also some post-translational modification information. Due to the large number of atoms in each protein molecule, the isotopic distributions of intact proteins are quite different from that of small molecules. This isotopic distribution causes much wider protein peaks. In this study, we investigated the affects of mass spectra deconvolutions on the mass accuracy and quantification of intact protein LC/MS analysis.
