IMS-MS was combined with supervised PCA analysis in order to analyze glycans isolated from serum of healthy individuals and patients with liver cancer and cirrhosis. In favorable cases, PCA of ion mobility profiles of single m/zion can be sufficient to cluster individuals into cohorts with specific physiologies, but combination of IMS profiles of multiple glycans separates better individuals with liver cancer and cirrhosis from the healthy group. Although supervised PCA uses prior knowledge of the sample groups, it can be useful test for analysis of unknown samples. For example, supervised PCA can be used to determine a linear discrimination function (LDF) for particular sample groups, and the LDF can be further calculated for the unknown samples. We plan on analyzing some of the unknown samples since this could be a final test for the use of proposed methodology for disease diagnosis.
