Milk has served as the primary source of nutrition for infants for millions of years due to its ideal composition of essential macromolecules, some of the most important being proteins. These biologically active proteins exert many essential functions, which include antimicrobial and immunomodulatory protection. In recent years there has been great focus on the milk proteome, and proteomic analysis of milk has made great strides, with the identification of over a thousand unique, and previously unknown, proteins being discovered. With the identification of so many unique proteins in milk, focus has turned to identifying specific proteins that may play a role in disease, either in the mother or infant. In this study we focused on proteins that are differentially expressed in gestational diabetes mellitus (GDM). GDM is a condition that affects between 3-10% of woman during pregnancy, yet very little is known about what roles it plays in lactation and milk protein content. Using a streamlined LC-MS/MS based approach, we were able to identify 601 proteins in human whey colostrum, 56 of which were shown to be significantly different between subjects with and without GDM.
