8,5′-cyclopurine-2′-deoxynucleosides (cPus) are formed in DNA upon exposure to hydroxyl radicals. cPus can block DNA replication and transcription, causing replication and transcriptional mutagenesis, and are considered reliable biomarkers of oxidative stress and oxidatively induced DNA damage. With the increasing demand for sensitive and rapid quantification of cPus in vivo, we are in the process of developing a nanoflow LC-MS/MS-based method for the quantitative measurement of cPus using a few micrograms of DNA samples that does not require the labor-intensive off-line HPLC enrichment procedures.
