CrkRS (Cdc2-related kinase, Arginine/Serine) or CDK12 (Cyclin Dependent Kinase 12) is a large serine/threonine kinase whose features suggests it may coordinately regulate RNA splicing and transcription (Ko, T.K. et al. J. Cell Sci.114: 2591-2600 (2001)). CrkRS has a N-terminal RS domain suggestive of a role in RNA splicing and has been found to associate with SC35 spliceosome-related nuclear speckles. Its kinase domain is closely related to several cyclin dependent kinases. While CrkRS complexes were known to phosphorylate the C-Terminal Domain (CTD) of RNA Polymerase II in vitro (Ko, T.K. et al. J. Cell Sci.114: 2591-2600 (2001)), the responsible cyclin was not known. However, previous studies have shown that the co-expression of CrkRS and Cyclin L, a cyclin which contains a RS domain, could affect the alternative splicing of a model substrate (Chen, H.H. et al. MCB 26(7): 2736-2745 (2006)).
