The poor accessibility to phosphoproteins notably low abundant ones gets researchers into trouble when looking for signalling components. With the establishment of this two step procedure (PAPE) we were able to highly enrich phosphor-proteins from complex leaf sample of Arabidopsis thaliana. The prefractionation step depleted almost all of the RuBisCO content and served as a first enrichment. Subsequent MOAC further increased the phosphoprotein content. Reproducibility could be visibly verified with a phosphospecific dye and more importantly via mass spectrometry. The detection of low abundant phosphoproteins, namely MPK6 and MPK4, could be achieved. The implementation of this method into our workflow of the artificial response system recently started and shows promising results. Future studies will focus on seeking for new MAPK3/6 targets with the help of the above system and the target confirmation.
