To investigate the mechanism underlying processing and release of incretins from enteroendocrine (EE) cells in the intestine, we performed a differential mass spectrometry (dMS) study to profile secreted peptide hormones from murine EE cell lines, GLUTag and STC-1, under basal and stimulated conditions. Secreted peptidomes in the cell media under the two conditions were analyzed on a nanoflow LC-FTMS platform; only peptides with elevated levels under stimulated condition were subjected to sequencing. Two important features of this approach, a system free of intracellular protein background and selection of precursor ions of secreted peptides, facilitated an efficient method for cataloging bioactive peptides. Consequently, high sequence coverage of several key prohormone sequences was achieved, providing insights into regulated hormone secretion from EE cells.
