Flow Cytometric Identification of Macrophages and Polymorphonuclear Leukocytes in Bovine Milk

The macrophages are the main leukocyte present in healthy mammary glands (Sarikaya et al. 2004). Once invading pathogens are detected, the resident macrophages release cytokines, eicosanoids, acute phase proteins and chemoattractants that direct migration of polymorphonuclear leukocytes (PMNLs) from the blood to the site of infection and consequently lead to an increase in milk somatic cell count (SGC). The flow cytometry is an easy and fast way to identify each population that can be use to diagnosis intramammary infections (IMI), even in the early stages of infection. In contrast, the differential inflammatory cell count by microscopy method consumes much time and also has poor repeatability (Koess and Hamman, 2008). Indeed, it is the first report, to our knowledge, that described the expression of CD 14 in PMNLs from bovine milk using specific monoclonal antibodies by flow cytometry. Thus, the aim of the present study was to provide a flow cytometry procedure for correctly identification of macrophages and PMNLs in bovine milk.
