To explore the contribution of impaired red blood cell (RBC) flow properties to circulatory disorders, RBC were treated with a mild concentration of hydrogen peroxide (HPO) and their deformability and adhesion to endothelial cells (EC) were measured in vitro. In parallel, the HPO-treated RBC were perfused into rat mesocecum vasculature, and vascular resistance was monitored. HPO-induced increase in vascular resistance was correlated primarily with increased change in RBC/EC adhesion. The results of this study clearly show that mild treatment of RBC with HPO (0.5 mM) affects, primarily and markedly, their adherence to EC of vessel wall, thus demonstrating the special sensitivity of RBC adherence to OS states.
