Serum levels of TNF-a and IL-18 correspond to severity of disease during systemic inflammation. SNPs affect the expression of these cytokines and show ethnic differences. This study aimed to determine the relations between the expression and SNPs of these cytokines in Japanese subjects. Blood was obtained from healthy volunteers to isolate DNA and PBMCs. Promoter SNPs were determined by sequence-specific PCR. Isolated PBMCs were cultured for 24 h with or without LPS, and cytokine levels were measured by ELISA. The TNF-a levels for the TNF-a -238 GA genotype were significantly lower than that for the TNF-a -238 GG genotype. IL-18 production did not differ among IL-18 -607 genotypes. The TNF-a -238 GA genotype may have low TNF-a production and IL-18 -607 polymorphism may not affect the production of IL-18 in Japanese.
