ASABF-α (Ascaris suum antibacterial factor) susceptibility is variable among S. aureus strains, but its mechanism is still elusive. In our analysis examining the relevance of the phospholipid composition to the ASABF-α resistance, lower amounts of cardiolipin (CL) were detected from the ASABF-α resistant strains probably due to their thick cell walls that interfered with the extraction of CL. Indeed, when the cell wall was destructed by lysostaphin, the extracted CL from each strain was indistinguishable irrespective of the ASABF-α resistance. Furthermore, the deletion of two CL synthase genes did not affect the ASABF-α susceptibility. Taken together, the physical characteristics of the cell surface, rather than the CL content, was supported as an important factor interfering with the ASABF-α activity.
