Outer membrane vesicles (OMV) are released by evaginations of the outer membrane of Gram negative bacteria and have gained attention with respect to their biological functions, role in pathogenesis and use in new vaccines strategies and combinations. OMV from Neisseria lactamica induce antibodies that cross-react with Neisseria meningitidis and can have a potential mucosal adjuvant property. The objective of this work was to optimize OMV production from N. lactamica through the modification and improvement of its original culture medium, meningococcus defined medium (MCDA). Experiments were carried out in batch processes which had their concentration of lactate, amino acids and yeast extract (YE) modified according to each essay. Results indicated that the use of threefold of MCDA's concentration of lactate and double of its concentration of amino acids plus 2 g/l of YE seems to be the most favourable condition in both terms of cost and productivity of OMV. This proportion between carbon and nitrogen sources appears to be the most adequate for N. lactamica's metabolism.
