To study the pharmacokinetic parameters of Peimine and Peiminine in rat plasma after oral administration of Danggui-Beimu-Kushen-Pill(DBKP). The analytes were extracted by liquid-liquid extract with diethyl ether and sinomenine was used as internal standard. Chromatographic separation was achieved on a Atlantis T3 column using a gradient elution program with methanol and water containing 10 rnmol/L ammonium acetate. The detection was performed on a triple-quadrupole tandem mass spectrometer by multiple reactions monitoring (MRM) mode in a positive ion mode via electrospray ionization (ESI). Linear calibration curves were obtained in the concentration range of 0.2-25 ng/mL and 0.4-50 ng/mL for peimine and peiminine. Extraction recovery was over 80% in plasma. The intra- and inter-day precision and accuracy were below 10% at three QC levels for both analytes. The method was shown to be effective, convenient, and suitable for simultaneous pharmacokinetic study of peimine and peiminine, which would be beneficial for the clinical use of DBKP.
