Carp which received intraperitioneal injection of sodium alginate showed high survival rate after challenge with E. tarda. In order to elucidate the immunoen-hancement by sodium alginate, we investigated its effects on the non-specific immune system of caro. Sodium alginate gave little effect in vivo and in vitro on the alternative complement pathway of carp. Intraperitoneal injection of alginate (2 g/100 g body wieght) did not affect the phagocytic activity of head kidney phagocyts (HKP), but greatly enhanced the migration of phagocytes to the peritoneal cavity. The number of induced cells was 16 to 50 times greaer than other kleukocyte-inducing substances (casein sodium, glycogen, liquid paraffin. polypeptone-soluble starch (1:1)and sodium thioglycollate) when injected at the same dose. In chemotaxis assays, migration of normal HKP to the culture supernatant of peritoneal exudate cells (PEC) increased when PEC were cultured with alginate, and migration of HKP obtained from alginate-injeted fish was significantly higher than that of normal HKP. These results indicate that sodium alginate stimulated PEC to produce chemotactic factor and concurrently enhanced chemotactic activity of HKP.
