Abstract: In the body, individual T helper lymphocytes need to beactivated first by physical contact withantigen-presenting cells (APC). T-cell contact withAPCs initiates an activation cascade which includes anincrease in T-cell intracellular calcium and leads toT-cell proliferation and differentiation. We havecombined fluroescence spectroscopy and imaging withoptical manipulation to investigate the physicalproperties of T-cell activation. We study cell-cellcontact requirements for T-cell activation usingoptical tweezers to control the orientation ofT-cell/APC pairs and fluorescence microscopy to measurethe subsequent T-cell intracellular calcium level($LB@Ca$+2$PLU$/$RB$-i$/) response. APCs or beadscoated with antibodies to the T-cell receptor aretrapped with a near- infrared titanium-sapphire laserand placed at different locations along the T-cell,which has a polarized appearance defined by the shapeand direction of crawling. T cells which are presentedwith antigen at the leading edge have a higherprobability of responding and a shorter latency ofresponse than those contacting APCs or beads with theirtrailing end. Alterations in antibody density and beadsize are used to determine the spatial requirements forT cell activation and the minimum number of receptorswhich must be engaged in order to transmit a positivesignal. !0
